DNA hasar\u0131 (genotoksisite) de\u011ferlendirmesi bu test yakla\u015f\u0131m\u0131n\u0131n temel dayana\u011f\u0131d\u0131r. G\u00fcn\u00fcm\u00fczde ve ge\u00e7mi\u015fte ger\u00e7ekle\u015ftirilen \u00e7o\u011fu in vitro test do\u011fas\u0131 gere\u011fi akut uygulanmakta, y\u00fcksek konsantrasyondaki test ajan\u0131 tek seferde h\u00fccre ortam\u0131na verilmekte ve daha sonra DNA \u00fczerindeki etkisi belirlenmektedir. Bu y\u00f6ntem, d\u00fc\u015f\u00fck doz ve kronik olarak ger\u00e7ekle\u015fen insan maruziyetlerinden tamamen farkl\u0131d\u0131r. Akut dozlama h\u00fccresel savunmalar\u0131 bertaraf etme ve yanl\u0131\u015f pozitif sonu\u00e7 verme yetene\u011fine sahiptir. Elde edilen baz\u0131 veriler kronik dozlaman\u0131n daha az genotoksik yan\u0131t olu\u015fturdu\u011funu ve insan maruziyet senaryolar\u0131yla daha uyumlu oldu\u011funu g\u00f6stermektedir. 13-cis-retinoik asitin gebelik kategorisi \u201cX\u201d olup, Grup 1B\u2019de s\u0131n\u0131fland\u0131r\u0131lmaktad\u0131r. Akne vulgaris tedavisinde g\u00fc\u00e7l\u00fc etkinli\u011fiyle di\u011fer tedavi y\u00f6ntemlerine yan\u0131t vermeyen \u015fiddetli akne durumunda tercih edilen bir ila\u00e7t\u0131r. Teratojenik etkinli\u011fi kan\u0131tlanm\u0131\u015f olmakla birlikte, mutajenik ve karsinojenik etkinli\u011fi ile endi\u015fe yaratmaktad\u0131r. 13-cis-retinoik asit\u2019e maruziyetin reaktif oksijen t\u00fcrlerinin art\u0131\u015f\u0131na, antioksidan kapasitenin zarar g\u00f6rmesine neden oldu\u011funu bildiren \u00e7ok say\u0131da ara\u015ft\u0131rma bulunmakla birlikte, sitotoksik ve genotoksik etkinli\u011fi tart\u0131\u015fmal\u0131d\u0131r. 13-cis-retinoik asit\u2019in g\u00fcn\u00fcm\u00fczde de akne vulgaris tedavisinde yayg\u0131n kullan\u0131m\u0131 ve tedavide ba\u015far\u0131l\u0131 sonu\u00e7lar\u0131n elde edilmesi maruziyet riskinin artmas\u0131na neden olmakta, bu y\u00f6n\u00fcyle de konu halk sa\u011fl\u0131\u011f\u0131n\u0131 yak\u0131ndan ilgilendirmektedir. Bununla birlikte literat\u00fcrde kronik dozlaman\u0131n akut dozlama ile kar\u015f\u0131la\u015ft\u0131r\u0131lmas\u0131na ve sitotoksik-genotoksik etkinli\u011fe p-53 etkisinin ara\u015ft\u0131r\u0131lmas\u0131n\u0131 da i\u00e7eren bir in vitro temelli ara\u015ft\u0131rmaya rastlan\u0131lmam\u0131\u015ft\u0131r. Ara\u015ft\u0131rmam\u0131zda in vitro karsinojenite testlerinin insanlar\u0131n ger\u00e7ek maruziyet ko\u015fullar\u0131na benzer olmas\u0131 amac\u0131yla, 13-cis-retinoik asit, TK6 ve NH32 (P53 geninden yoksun) gibi \u00f6zel lenfoblastoid h\u00fccrelere optimize edilecek doz rejiimleriyle akut ve kronik dozlarla uygulanacak, h\u00fccrelerde olu\u015fturaca\u011f\u0131 genotoksik hasar MN testiyle, sitotoksik hasar ise relatif population doubling (%RPD) testiyle saptanacakt\u0131r. TK6 ve NH32 h\u00fccrelerindeki apoptoz mekanizmas\u0131n\u0131n farkl\u0131l\u0131\u011f\u0131n\u0131n sitotoksisite ve MN sonu\u00e7lar\u0131na olan etkinli\u011fi de\u011ferlendirilecektir. Kimyasal maddelerin h\u00fccre hatlar\u0131nda ortaya \u00e7\u0131kard\u0131klar\u0131 sitotoksik ve genotoksik etkinlikler maddelerin toksik etki mekanizmalar\u0131na ba\u011fl\u0131 olarak farkl\u0131l\u0131k g\u00f6sterebilmektedir. Bu \u00e7al\u0131\u015fmada amac\u0131m\u0131z in vitro deney \u015fartlar\u0131nda 13-cisretinoik asit\u2019in TK6 ve NH32 h\u00fccreleri \u00fczerinde ortaya \u00e7\u0131kartaca\u011f\u0131 sitotoksik ve genotoksik hasar\u0131n s\u0131ras\u0131yla %RPD ve CB-MN y\u00f6ntemleri ile belirlemek, akut (1+2 g\u00fcn) ve kronik (5+2 g\u00fcn ve 10+2 g\u00fcn) dozlama uygulamalar\u0131n\u0131n onun toksik etkinliklerine ne y\u00f6nde etki yapt\u0131\u011f\u0131n\u0131 ve p53 geninin ortaya \u00e7\u0131kabilecek karsinojenik etkiye olan etkisini ara\u015ft\u0131rmakt\u0131r.<\/p>\n<p><strong>Kifayet S\u00f6nmez<\/strong>\u2019in, \u201cSunset Yellow\u2019un in vitro genotoksik akut ve kronik dozmaya ba\u011fl\u0131 p53 geni etkinli\u011finin mikro\u00e7ekirdek y\u00f6ntemi ile de\u011ferlendirilmesi\u201d adl\u0131 projesinde ise, Sunset yellow sentetik anyonik bir azo boyas\u0131d\u0131r. Turuncu rengin farkl\u0131 tonlar\u0131n\u0131 verebilmesi s\u00fcre ve s\u0131cakl\u0131\u011fa ba\u011fl\u0131 kal\u0131c\u0131l\u0131\u011f\u0131, stabilitesinin y\u00fcksek olmas\u0131 nedeniyle \u00e7e\u015fitli g\u0131da \u00fcr\u00fcnlerinde, parf\u00fcmeride, hatta farmas\u00f6tik preparatlarda yayg\u0131n olarak tercih edilmektedir. Baz\u0131 Avrupa Birli\u011fi \u00fclkelerinde kullan\u0131m\u0131 s\u0131n\u0131rland\u0131r\u0131lm\u0131\u015f ve yasaklanm\u0131\u015ft\u0131r. Mutajenik ve karsinojenik etkinli\u011fi tart\u0131\u015fmal\u0131 olmakla birlikte baz\u0131 \u00e7al\u0131\u015fmalarda sitotoksik ve genotoksik etkinliklerine ek olarak haz\u0131r g\u0131da \u00fcr\u00fcnlerinde yayg\u0131n kullan\u0131mlar\u0131 nedeniyle halk sa\u011fl\u0131\u011f\u0131 a\u00e7\u0131s\u0131ndan endi\u015fe yaratmaktad\u0131r. Maruziyetinin reaktif oksijen t\u00fcrlerinin art\u0131\u015f\u0131na neden oldu\u011funu bildiren \u00e7ok say\u0131da ara\u015ft\u0131rma bulunmakla birlikte, literat\u00fcrde kronik dozlama maruziyete dayal\u0131 in vivo \u00e7al\u0131\u015fmalar bulunmas\u0131na kar\u015f\u0131n, kronik dozlamaya dayal\u0131 in vitro bir ara\u015ft\u0131rmaya rastlan\u0131lmam\u0131\u015ft\u0131r. Ara\u015ft\u0131rmam\u0131zda in vitro karsinojenite testlerinin insanlar\u0131n ger\u00e7ek maruziyet ko\u015fullar\u0131na benzer olmas\u0131 amac\u0131yla, sunset yellow, TK6 ve NH32 (P53 geninden yoksun) gibi \u00f6zel lenfoblastoid h\u00fccrelere optimize edilecek doz rejiimleriyle akut ve kronik dozlarla uygulanacak, h\u00fccrelerde olu\u015fturaca\u011f\u0131 genotoksik hasar MN testiyle, sitotoksik hasar ise relatif population doubling (%RPD) testiyle ara\u015ft\u0131r\u0131lacakt\u0131r. Konuyla ilgili yapt\u0131\u011f\u0131m\u0131z \u00e7al\u0131\u015fmalar genotoksik kimyasallar\u0131n NH32 h\u00fccrelerinde daha fazla MN hasar\u0131 olu\u015fturmas\u0131na kar\u015f\u0131n, TK6 ile kar\u015f\u0131la\u015ft\u0131r\u0131l\u0131nca bu h\u00fccrelerin daha fazla canl\u0131 kald\u0131klar\u0131n\u0131 g\u00f6stermektedir. Yine elde etti\u011fimiz veriler, ayn\u0131 deney \u015fartlar\u0131nda kronik dozlama uygulamas\u0131n\u0131n baz\u0131 kimyasallarla (H2O2 ve menadion) daha az MN hasar\u0131 olu\u015fturmas\u0131na kar\u015f\u0131n, baz\u0131 kimyasallar\u0131n (KBrO3) kronik dozlama uygulamas\u0131yla daha fazla MN hasar\u0131na neden oldu\u011fu g\u00f6stermi\u015ftir. Sonu\u00e7 olarak akut doz uygulamas\u0131 kronik dozlamaya g\u00f6re beklendi\u011fi gibi her zaman daha fazla genotoksik ve sitotoksik hasar olu\u015fturmamaktad\u0131r. Kimyasal maddelerin h\u00fccre hatlar\u0131nda ortaya \u00e7\u0131kartt\u0131klar\u0131 sitotoksik ve genotoksik etkinlikler maddelerin toksik etki mekanizmalar\u0131na ba\u011fl\u0131 olarak farkl\u0131l\u0131k g\u00f6sterebilmektedir.
